Human T cells are now relatively easy to modify ex vivo, but mouse T cells are a bit finicky. We made a resource in Nature Protocols for CRISPR-editing of primary mouse T cells, hoping to establish a pipeline for generating knockout CAR T cells for use in syngeneic models.
