Interesting preprint about the proportional predictive value of PRSs for CVD being way higher in ages <55y, where clinical risk factors are rarer and carry less prognostic power medrxiv.org/content/10.110… Similar data previously for other disease outcomes journals.plos.org/plosgenetics/a…

Great to showcase this work on lamin heart disease at #AHA23. Subclinical #LMNA variant carriers may have worse myocardial mechanics, higher T2, higher ECV, and more LGE compared to healthy volunteers. Gabriella Captur; James moon LHA @ UCL/UCL Institute of Cardiovascular Science; Barts Health

Nothing makes me as uneasy as a human geneticist as seeing the proliferation of direct to consumer polygenic risk score companies. I know academics are conservative but these scores are truly not good enough yet & have potential for true harms

Examining the cross-ancestry generalizability of polygenic risk scores, Human Genetics and Genomics Advances' latest study discovered that PGS313 predicts breast cancer risk, but with reduced accuracy, for American women of African, Asian, and Latinx ancestry: cell.com/hgg-advances/f… Bogdan Pasaniuc #ASHG

Really nice method for computing context specific polygenic score uncertainty. PRS methods are so far ahead of most clinical scores on this point. nature.com/articles/s4158…

Do the multiple existing PRSs offer reliable individual-level assessment of disease risk❓ 👉48 PRSs for coronary artery disease 👉comparable population-level calibration and discrimination ❗️wide variance in individual-level risk depending on PRS 🔗medrxiv.org/content/10.110…

Despite similar performance at the population level, different coronary heart disease polygenic risk scores produced highly variable individual-level risk estimates. ja.ma/42gmKCu
