To optimize RESTRICT-seq without relying on extensive, time-consuming knockout assays for every condition, we developed the arCas9 Modeling for Activity Prediction (arMAP) algorithm. This revealed that nuclear localization of Cas9 serves as a direct proxy for its DNA cleavage.
