This study started with a question: what are the unknowns of translational control in breast cancer metastasis? By combining Ribo-seq and Ribolog, a new tool by Hossein Asgharian for Ribo-seq analysis, we found HNRNPC to control translation of a metastasis suppressing mRNA regulon.

HNRNPC is known as a nuclear regulator of alternative splicing – how could it regulate translation? It turns out HNRNPC interacts with PABPC4, a poorly characterized poly(A) binding protein, and binds target mRNAs near poly(A) signals.

In highly metastatic cells, the lower expression of HNRNPC and PABPC4 results in the 3' UTR lengthening of their target mRNAs and subsequent translational repression, at least partly through an Argonaute-dependent RNAi.

Lower expression of the HNRNPC-PABPC4 axis components increases metastasis in xenograft mouse models and is associated with poor clinical outcomes in breast cancer patients.

Finally, modulating the poly(A) site selection using a small molecule drug reduces the metastatic lung colonization, providing evidence for a therapeutic vulnerability of HNRNPC-low tumors.
