The crystal structures are of a different protein construct than what is used in the affinity expts. The crystals are missing a regulatory domain. The binding site in the crystal is half solvent exposed with picomolar ligands bound. Odd!

Your future research plan is the most important part of the application. Show them that you want to do exciting research that really moves the needle in your field. Also, having a high number of papers published gets you noticed.

Very nice work. We had a paper on the Biophysical Limits of Protein-Ligand Binding back in 2012 where we showed the highest LE we’re charged ligs in charged pockets… biotin is definitely a different beast in the lig universe!! Thanks for highlighting it.
