Excited to share our total synthesis of aporphine alkaloids! Inspired by biosynthesis, we use a non-natural intermolecular photochemical phenol coupling to access analogues for serotonin & adrenergic receptor profiling. Kozlowski Lab Total Synthesis pubs.acs.org/doi/10.1021/ac…

Rowan With glaucine and methylisothebaine in hand, we synthesized a variety of chemically diverse analogues. We tested these compounds at the serotonin 5-HT2 and adrenergeric alpha-1A receptors through the National Institute of Mental Health (NIMH) PDSP program.

Kalyana Duggal analyzed all of the biological data using machine learning to help rationalize the binding affinity across the alkaloid chemical space. RDKit-chemical descriptors enabled the development of a moderate linear regression model for the alpha-1A receptor activity.
