What if you could point to a brain region and measure c-Fos, or assess the success of gene delivery? We combined synthetic markers with focused ultrasound to enable site-specific measurement of gene expression in the brain. More in the preprint: biorxiv.org/content/10.110…

Many brain therapies are permanent. One can't undo surgical resection for epilepsy, or remove gene therapy from the brain. What if we could do a test drive? RAID is a new concept for non-genetic, temporary, localized pharmacological control of the brain. biorxiv.org/content/10.110…

Serum markers are very useful, but they can't track many processes in the body. What if we could make synthetic serum markers to track any process? What if we could measure gene delivery or neuronal activity with a blood test? see our paper Nature Biotechnology nature.com/articles/s4158…

Can AAVs be evolved for better brain delivery after targeted ultrasound insonation? We report new AAVs optimized for focused ultrasound BBB opening. Congratulations to all involved! Richard Hongyi Li Manwal Harb John Heath @JakeSTrippett Mikhail Shapiro (same on bsky) 🇺🇦 nature.com/articles/s4146…

Thrilled to announce that I have accepted a TT faculty position at the University of Pittsburgh in Pitt Neurosurgery, starting 01/2025! We will develop noninvasive molecular tools to control and monitor endogenous processes in intact cells and tissues, focusing on the brain. 1/2

How could we check if a gene therapy reached the desired brain region? Here, we noninvasively measure gene delivery and endogenous gene expression in specific brain sites with ultrasound-released synthetic serum markers. science.org/doi/10.1126/sc…

qPCR can measure expression of virtually any gene... in homogenized tissues. Here, we show an "in-vivo qPCR" equivalent, where we measure a sequence-specific transcript with synthetic serum markers and enzyme-based signal amplification. biorxiv.org/content/10.110…

Our preprint is out! We developed a new technology that enables noninvasive monitoring of mRNA expression. Imagine measuring the mRNA expression as easy as qPCR, but in living animals - it’s now a reality. biorxiv.org/content/10.110…

Our outgoing postdoc Zhimin Huang starts his lab Pitt Neurosurgery January 1st, but already got a PhRMA foundation Faculty Starter Grant to develop site-specific brain therapeutics. I'd bet my money on this being just the first of many awards. phrmafoundation.org/news-events/pr…

We have two open postdoc positions, focused on molecular engineering, viral vector development, and a large animal focused ultrasound. It's a 4-year long, UK gov funded study. We will be looking at the candidates in mid-late February. please RT. emdz.fa.us2.oraclecloud.com/hcmUI/Candidat…

A new Ph.D from our lab! Congrats to Dr. Joon Pyung Seo. Dr. Seo established REMIS for measuring gene expression in the brain with spatial precision, contributed to the developing site-specific brain therapeutics, and enabled a clinical trial through his data. Links in comments

Excited to talk about how we used a retrograde-tracing viral vector for noninvasive transduction of neuronal projections at my upcoming defense! Grateful for the support of my lab mate Shirin Nouraein and my advisor Jerzy Szablowski.

PhD #3 - Dr. Manwal Harb who helped to develop the first engineered AAV for focused ultrasound delivery, evaluated a retrograde-tracing AAV9 for noninvasive targeting of neuronal projections, and contributed to other projects. He will move on to be a CEO of a stealth startup.

Usually serum markers are detected as they are - often hard to detect, long half-life, or with high background that reduces temporal resolution. What if we could modify the markers in vivo so they can better match our needs? Introducing this concept with erasable serum markers.

Excited to share my recent work on erasable synthetic serum markers-engineered RMAs with a controllable half-life to reduce background noise and enable sensitive, high-temporal-resolution monitoring of brain gene expression, all with a simple blood test!🧠🩸
