4/8 🗺️ We believe that such false neg results are due to the low resolution of the cis-regulatory element (CRE) map. Using #singlecell #multiome, we built a genome-wide macrophage CRE map, linked 121 CAD risk variants to 56 genes, and revealed an AP-1-centered regulatory network!

7/8 🎯 Mechanistically, we have shown that the transcriptional regulatory machinery for a CAD risk variant, rs10488763, is specifically activated in lipid-handling macrophages. Also, for the first time, we have implicated its target FDX1 as a disease gene for atherosclerosis!
