Using the DUB mutant mice together with cell biology, XL-MS, and cryo-EM, we show that K63-Ub of A-complex substituents engages the RAP80 UIM domains. This prevents ubiquitin recognition at damage sites. DUB activity removes K63-Ub to license A-complex DDR function. 3 of 3.
