Here is a thread summary of our recent paper: Evolutionarily conserved enhancer-associated features within the MYEOV locus suggest a regulatory role for this non-coding DNA region in cancer doi.org/10.3389/fcell.… LJ Russell Lab Cabimer @NUBiosciences 1/7

The gene MYEOV (MYEloma OVerexpressed) was first described as a highly transcribed gene in myeloma, but it is over-expressed in many cancer types, and it has been proposed to have functions as RNA (unclear as protein). Protein-coding region is only conserved in primates. 2/7

MYEOV is located in the same TAD that CCND1. In our previous paper, Aneta Mikulasova noticed that MYEOV has enhancer chromatin features genome.cshlp.org/content/32/7/1… - This led to Brigid's MRes master project (linkedin.com/in/brigid-davi…) with Marco Trevisan-Herraz 🔭🦠🚲🌱🌍 LJ Russell Lab #JulianaArcila 3/7

Brigid found additional experimental evidence that suggests that a region overlapping MYEOV in humans and other primates could function as an enhancer in many cell types, has enhancer potential (STARR-seq) and physically interacts with CCND1 and LTO1 genes. 4/7

BLAST evolutionary analysis of this DNA region showed that the DNA sequence is deeply conserved in all mammals analysed, and even found in chicken (and always sits next to the CCND1 homologs). Helped by PhDstudent #JulianaArcila (linkedin.com/in/jearcilaga), joint first author. 5/7

And the homolog DNA region in mouse (no protein-coding region present) also shows physical interactions with mouse Ccnd1 gene. This opens the future possibility to study the potential enhancer function in mouse models. All results obtained with existing data, thanks to all! 6/7

Finally, I would like to thank co-author #ChrisBrackley for his work using HipHop polymer simulations confirming the interactions of the DNA region and CCND1 in humans. More about this approach here: genome.cshlp.org/content/32/7/1… Enjoy! 7/7

PS. I posted this thread on August 8th, probably the worst time of the year to announce a new paper and a thread summary... Hopefully I can bring more attention now most of us are "back to school" :-) LJ Russell Lab IHEC_Epigenomes Cabimer @NUBiosciences

Today we have Daniel Rico from Cabimer talking about chromatin structure (TADS), gene age and essentially. scholar.google.com/citations?view… Connected with our old paper “ Late-replicating CNVs as a source of new genes” Juan, Rico, et al Biology open 3, 231-231, 2013

Good news! Our article with Daniel Rico and Aneta Mikulasova and led by previous PhD student Daniel Kent and PDRA Letizia Marchetti received enough citations to be a Top Cited Article. Read it here onlinelibrary.wiley.com/doi/full/10.10… #TopCitedArticle Wiley in research
