Disappointingly, none of these papers are addressed in the Nature Cancer manuscript, and no effort is made to show that ricolinostat is specific for HDAC6. The prior literature and the potential safety concerns that they raise are just ignored.

Usually I let these things go by without commenting, but in this case, the authors conducted a clinical trial and gave this drug to patients. In light of that, I think that heightened scrutiny around safety/off-target toxicity concerns is very warranted.

To be clear - our results were conducted in cell line models and investigated the question of whether HDAC6 is required in a cell-autonomous manner for cancer growth. We do not rule out the possibility…

that HDAC6 inhibition has on-target effects in a cell non-autonomous process like inflammation. Still, that’s not what this paper claims, and the authors provide no evidence for an on-target, non cell-autonomous effect.
