Expression and purification of Tn5 protein in house has proven difficult for many labs, which limits both the equitable adoption of existing tagmentation methods, as well as hindering the development of novel ones. 4/11

In OpenTn5, we integrated and optimized the many existing resources into a unified, start-to-finish open-source protocol, to reliably produce large quantities of a multi-functional pG-Tn5 enzyme. 5/11

We benchmarked pG-Tn5 against commercial enzymes in both CUT&Tag as well as bulk ATAC-seq assays. pG-Tn5 is suitable for use in both of these applications; and the pG tag does not detectably interfere with even the very fine footprinting features of ATAC-seq data! 8/11

Because we store the pG-Tn5 stock unloaded, custom sequencing adapters can be easily used for different applications. We demonstrate this by applying pG-Tn5 to the recently developed single-cell EasySci-ATAC. 9/11
